Resumo
A Leucemia Mielóide Crônica (LMC) é um tipo de neoplasia caracterizada pela proliferação anormal e descontrolada, com progressiva perda de diferenciação, de células do tipo granulocíticas. A hiperatividade da tirosina quinase, fruto de mutagênese, característica base da doença, inibe a apoptose e estimula a mieloproliferação. A partir de 1993, com a sintetização do mesilato de imatinibe, inicia-se uma nova etapa no tratamento da LMC, através da terapia molecular, com a utilização de inibidores da enzima tirosina quinase. Esses fármacos inibem de forma seletiva a proliferação e induzem a apoptose em linhagens de células bcr-abl positivas, promovendo, se não a cura, o controle da doença, aumentando significativamente a expectativa de vida. Muito pacientes, porém, apresentam resistência ao imatinibe, ou evoluem com refratariedade ao tratamento. A biotecnologia tem papel de extrema importância na identificação das causas e na elucidação dos mecanismos envolvidos nesse processo, sendo ferramenta fundamental no desenvolvimento de novas gerações de inibidores de tirosina quinase. Através da farmacogenética, é possível a individualização do tratamento, de acordo com as características genéticas de cada paciente, seu metabolismo e mutações apresentadas, apontando assim, a opção terapêutica mais eficaz.

Abstract
Chronic Myeloid Leukemia (CML) is a type of neoplasm characterized by the abnormal and uncontrolled proliferation, with progressive loss of differentiation, of granulocytic type cells. The hyperactivity of the tyrosine kinase, the fruit of mutagenesis, the basic characteristic of the disease, inhibits apoptosis and stimulates myeloproliferation. From 1993, with the synthesis of imatinib mesylate, a new step in the treatment of CML begins, through molecular therapy, with the use of inhibitors of the enzyme tyrosine kinase. These drugs selectively inhibit proliferation and induce apoptosis in bcr-abl positive cell lines, promoting, if not cure, disease control, significantly increasing life expectancy. Many patients, however, are resistant to imatinib, or progressively refractory to treatment. Biotechnology plays an extremely important role in the identification of the causes and elucidation of the mechanisms involved in this process, being a fundamental tool in the development of new generations of tyrosine kinase inhibitors. Through pharmacogenetics, it is possible to individualize the treatment, according to the genetic characteristics of each patient, its metabolism and mutations presented, thus indicating the most effective therapeutic option.
